Determination of the presence of M. bovis in bovine tissue samples by RT-PCR

Abstract
Validation of the proof of RT-PCR to diagnose bovine tuberculosis caused by M. bovis was carried out. Doing the comparison against the results obtained previously by means of crop in half StonebrinkR. The aim was to evaluate the sensitivity and specificity of the proof and verify if this awards a greater proportion of true negative and true positive when diagnosing said illness unlike the proof of gold. 100 bovine (M. bovis) positive fabrics and 10 negative samples (M. bovis) were used; direct extraction of DNA by the method of CTAB was done. The protocol of RT-PCR and the oligonucleótidos reported by the United States Department of Agriculture (USDA) were used. Data were analyzed using the program EPIDAT 3.1. The results obtained had a sensitivity of 92% and a specificity of 100%, in an approximately 48 hours, DNA concentration was lower than 50 ng/µL.
